在T cell轉譯因子GATA-3刺激下,T lineage和Mast cell lineage之轉換

在第一眼瞥見T cell和mast cell時,大部分的人都會認為這是兩種個別不同的immune cell type。Mast cell屬於innate immune cell,大都留守在tissue中很少有機會進入血液,當antigen出現時,pattern-recognition receptor (PRR) 會刺激mast cell活化,引起立即且有效的proinflammatory response。相反地,T cell則是主要參與在adaptive immune response,平常在血液和淋巴結中循環,在經由antigen present cell將antigen呈現給T cell後,促使T cell活化並到達感染位置進行pathogen的撲殺,最後T cell會發展記憶性T cell,提供長時期的保護作用。
原本是差異很大的兩種immune cell,卻在本篇研究中拉起了線。在T cell progenitor進入thymus之後,正常情況下,stroma cell會傳遞Notch-Delta signaling給thymocyte使其進入下一階段的T cell differentiation。然而,倘若stroma cell無法持續傳遞Notch-Delta signaling,且將T cell早期發展必要的轉譯因子GATA-3大量表現時,會促使thymocytes表現高量c-Kit,並且使thymocyte走向mast cell differentiation。這樣的現象似乎在告訴我們:mast cell和T cell這兩個lineage在發育過程中有密切的關係存在。

就目前研究所知,我們知道mast cell源自bone marrow的hematopoietic stem cell,當bone marrow的stroma cell表現IL-3和SCF時,會使stem cell encode出GATA-1, GATA-2等等transcription factor,接著mast cell會表現tissue-specific homing molecule,使mast cell precursor travel 到target tissue,最後分化成熟。在T cell分面,hematopoietic cell會從bone marrow migrate到thymus中,經由Notch-Delta ligand 刺激以及GATA-3的調控,發育為成熟T cell。而本篇作者認為,當hematopoietic stem cell進入到thymus,分化到double negative 3 stage之前,這些thymocytes有可能因為其他Notch-Notch ligand signal,使thymocyte大量表現GATA-1, 2, 3,甚至bypass Notch-Delta signaling,最終分化成mast cell。
在發現的背後,還有一些問題仍需要去釐清。有些研究指出GATA-1, 2, 3都share一樣的DNA binding site,但是在in vivo情況下,GATA-3是否也會幫助mast cell differentiation?此外,mast cell是否真的可以在thymus中發育?如果可以,這些mast cell是會在thymus中成熟並且長久居留,還是最後會travel到其他target tissue?
