Induced recruitment of NK cells to lymph nodes provides IFN-γ for TH1 priming

Abstract

    Natural killer (NK) cells participate in innate and adaptive immunity, but the detailed mechanism in adaptive immunity is unclear1. NK cells can produce IFN-γ which plays a dual role in TH1 polarization2. It first increases the production of IL-12 by antigen-presenting dendritic cells (DCs). Second, it synergizes with T cell receptor (TCR) signals to induce the expression of T-bet, a transcription factor related to IL-12 receptor. In this study, the authors intent to find other cells to participate in T cell priming besides DCs. They demonstrated that NK cells could be recruited to lymph nodes after the injection of mature DCs into the mice. They also found that NK cells were recruited by the adhesion molecule CD62L (L-selectin) and the chemokine receptor CXCR3 rather than CCR7 in mice. In lymph nodes, NK cells could produce an early source of IFN-γ which stimulates TH1 polarization. Adjucvants are components of vaccines, which can induce cytokine release, the expression of costimulatory molecules and major histocompatibility complex (MHC), and upregulation of the chemokine receptor CCR7. The authors also found that some adjuvants, such as R848 and Ribi, could recruit NK cells to lymph nodes and induce TH1 polarization3. Taken together, NK cells migrated to lymph nodes via CXCR3 and participated in TH1 polarization by secreting IFN-γ. Furthermore, some adjuvants were also involved in the recruitment of NK cells.
