Abstract

A central doctrine of reproductive biology is that oocyte production in females of most mammalian species is believed to cease during fetal development1. In humans, exhaustion of the oocyte pool occurs around age 50 and the menopause comes. The authors show that adult female mice could rapidly generate oocytes in ovaries after oocytes are depleted by doxorubicin2. They also find that germ cells and bone marrow (BM) share similar markers. By transplanting the bone marrow from normal female mice intochemotherapy sterilized recipients, they find the oogenesis in adult female mice, indicating that a putative germline stem cell (GSC) reservoir resides in BM. If BM possesses GSC, the putative germ cells would travel to the ovaries for oocyte production by peripheral circulation. So the authors transfer peripheral blood (PB) harvested from transgenicfemales with green fluorescent protein (GFP) expression in germ cells into chemotherapy-treated female recipients, and the GFP-positive oocytes can be found in recipients. They have not yet determined whether the new generating oocytes can fertilize normally and produce viable offspring. The results demonstrate that BM and PB have potential sources of female germ cells and can sustain oocyte production in adulthood. This finding raises the hope for delaying the menopause and preserving fertility in female chemotherapy patients.
