TLR Signaling對於Salmonella typhimurium毒力影響是必要存在

摘要
Toll-like receptors (TLRs)會辨認各種不同微生物物質並啟動innate immune responses對抗外來微生物。然而,Salmonella typhimurium 可利用Salmonella pathogenicity island 2 (SPI-2)encode出的type 3 secretion system將SPI-2 effectors 注入宿主細胞將phagosomes轉化成適合複製的Salmonella-containing vacule (SCV)。但是什麼樣的訊息促使SPI-2表現則還不清楚。在這篇Paper中,作者發現TLR 2, 4 and 9 knockout (TLR 2x4x9 KO) mice相較TLR 2 and 4 knockout (TLR 2x4 KO) mice對於S. typhimurium敏感性較低(Fig. 1)。免疫螢光染色顯示S.typhimurium在TLR 2x4x9 KO bone marrow-derived macrophages (BMMs)的生長受到限制。Transmission EM data也發現S. typhimurium無法在TLR 2x4x9 KO BMMs形成SCV(Fig. 2)。此外,S. typhimurium在TLR 2x4x9 KO BMMs中也無法表現及secret對於intracellular growth 非常重要的SPI-2 effectors(Fig. 3)。最後他們證明TLR-dependent phagosomal酸化過程對於SPI-2 genes expression是必要的(Fig. 4)。這篇研究主要的發現為S. typhimurium利用TLR signaling調控virulence genes來達到一個成功的系統性感染。
