MED23 Mutation Links Intellectual Disability to Dysregulation of Immediate Early Gene Expression

摘要

真核生物的activator藉由兩個general mechanism會去調控轉錄作用。第一個是:Activator會與coactivator complex interaction後chromatin的結構會modified。general transcription factors(GTFs)和RNA polymerase II進入到DNA promoter中,接著transcrition initiation。 第二個則是:activator會促使preinitiation complex(PIC)(由general transcription factors+polymerase II)在promoter上形成,再刺激Pol II進行initiation。

Mediator complex在轉錄上扮演的角色是參與在transcriptional activation的第二個aspect和維持chromatin的hyperacetylated,將chromatin的結構打開。Mammalian mediator(MED)為一個multiprotein,其中MED23為其中的一個subunit。藉由genetic analysis去分析intellectual disability的病人的細胞,發現其MED23在cDNA 1850位置上有一個G to A的突變,amino acid會從    arginie轉變成glutamine。

根據以往的研究,MED23會去midiate immediate early genes (IEGs),

例如EGR-1, EGR-2, C-FOS and JUN,這些基因經過serum induction之後,會立即短暫的表現量改變。作者利用microarray的方式,將正常的細胞與帶有突變的細胞(M23/R617Q)經過serum free medium培養24小時之後,加入serum培養後,利用real-time PCR定量,比較兩著之間serum response genes的差異,觀察到JUN down regulation,FOS up regulation 的現象。

進一步為了去證找出是否這兩個IEGs在wild type與mutnat之間的表現量的差異,是由於MED23突變,而造成transcription調控異常所造成的。作者針對mediator的不同功能作探討:

另外,在轉錄的過程中,因為activator會recruit mediator使的轉錄進行。

 

所以作者檢視JUN與FOS promoter上activators的binding sites,利用chIP assay比較 wild type與M23/R617Q5 binding情形:

(1)在JUN promoter

當MED23 mutation之後,因為無法與Ternary complex factor(TCF)結合,chromatin loop無法產生,activator無法作用於JUN promoter,造成JUN表現下降。

 

(2) 在FOS promoter

當MED23 mutation之後,MED23會與ELK3結合,而不是與ELK1結合,造成FOS的表現量上升。

 

接下來,作者想知道是否因為mediator其他subunits mutation所造成的其他intellectual disability,是否也有IEGs表現量異常的情況。

可以看到也出現異常的情形,證明JUN與FOS在腦部的發育十分重要。

 

由以上結果:

作者證實IEGs的transcription與nonsyndromic intellectual disability的相關性。是否是因為IEGs表現異常造成shor-term神經活性改變,而產生此疾病,需要再加以研究。
