Plasmacytoid dendritic cells (pDCs) 具有Toll-like receptor (TLR)-7和 TLR-9可以偵測病毒的RNA與DNA;當病毒入侵時,pDCs可以釋放大量type 1 interferon並進一步來促進 B cells, T cells, natural killer cells 和 myeloid DCs的功能,啟動免疫反應來清除病毒;所以pDCs又被稱作 type 1 interferon-producing cells(IPC)。正常狀況下pDCs並不會偵測自體的DNA,但在某些自體免疫疾病如紅斑性狼瘡,pDCs會辨識antoantibody與自體DNA所形成的復合物進而被活化。而在乾癬,也是個常見的自體免疫疾病導致皮膚長期發炎,也可以觀察到pDCs進入患病組織中並釋放inferferon。另外作者發現到患處會釋放大量 LL37,一般被認為是 anti-microbial peptide 。但進一步實驗證明LL37會和DNA結合並使自體DNA聚集並濃縮,形成的復合物可進入 pDCs中並停留在early endosome的位置,進一步可以刺激位於early endosome的TLR-9,在訊息傳遞下可以使得pDCs表現並釋放大量IFNα,可以促進其他免疫細胞的活化並在患處造成自體免疫反應;作者認為這就是造成乾癬主要的治病機制,未來或許可以利用LL37的抑制劑來治療乾癬或是相關的長期發炎性疾病。
