Proteome analysis of soluble nuclear proteins reveals that HMGB1/2 suppress genotoxic stress in polyglutamine diseases

    Polyglutamine (PolyQ) diseases包含九種漸進式的遺傳性神經退化疾病,以Dr. George Huntington在1872年發表的Huntington’s disease(HD)為例,正常人的huntinntin gene具有6~34個不等的CAG三核甘酸重複序列。當此重複序列不正常增加,麩胺酸(Q)大量擴增於其蛋白質產物中,造成結構變異並形成包涵體(inclusion body)堆積在神經細胞核中,影響核內其它蛋白質的功能,損害synaptic recycling,患者隨著病程發展而產生中樞神經失調症狀(舞蹈症)。甚者因genotoxic stress導致神經細胞死亡。

    本文作者藉由蛋白質體學方法比較正常與不正常神經細胞核內的protein profiling,發現high mobility group B(HMGB, nuclear protein conserved across species, related to DNA recombination, replication and repair)與polyQ pathology相關,進而利用一連串的實驗證實mutant polyQ protein抑制HMGB表現量,導致genotoxic stress不能被HMGB消除。

    

 

這項研究為至今仍無法治癒的polyQ disease提供一個治療的目標。必須注意的是,外給HMGB會引起發炎反應2。如何將HMGB只送進細胞核中,將會是首要解決的問題。
